Viewers were left in suspense for multiple minutes while the race waited to get underway, with Lyles even being seen giving a signal to get organizers to hurry up.
It appeared that someone tried to run towards the track, causing the suspenseful delay, but the 100m final eventually got underway.
Lyles got out of the blocks slow, but his top speed helped him overcome Thompson at the line.
The 27-year-old finished the race with a time of 9.79 seconds, the fastest time of his career.
He was four thousandths of a second ahead of Thompson, who finished with almost the exact same time.
Michael Johnson, who commentated on the Olympic event for BBC, shared it was the greatest 100m he ever watched.
“Absolutely [the best 100m final I have seen] bar none. It was so open, there were so many great athletes,” he said.
“It lived up to it, we had this amazing race where you could throw a blanket over the finishing line.
“We didn’t even know who won for a few minutes.”
Lyles exploded with excitement after he was announced as the winner in the stadium and charged towards the camera to make a statement to the world.
“I told you!” he yelled to the world.
He was also seen sharing a touching moment with his mom after the race.
“I’m so proud of you,” Lyles’ mom repeated over and over again to her son.
Lyles repeatedly claimed himself as the fastest man in the world, and delivered on his promise in stunning fashion.
“It’s me. It’s always going to be me,” Lyles said of the fastest-man debate before the Olympics.
“Everybody knows that the title goes to the Olympic champion and the world champion, which I am one of, and soon to be another one of.
“A lot of people look at my confidence and… the line between confidence and cockiness is so blurred, it’s all an opinion.
